Sonogashira偶聯反應

背景介紹

1975年，K. Sonogashira和同事發現在溫和的條件下，乙炔氣與芳基碘化物或溴化乙烯反應，在催化量為Pd(PPh3)Cl2和碘化亞銅的存在下，可以制備對稱取代的炔烴。同年，R.F. Heck和L. Cassar的研究小組各自公開了類似的只使用Pd催化反應，沒有銅的協同催化，但反應條件苛刻。目前Sonogashira反應在取代炔烴以及大共軛炔烴的合成中得到了廣泛的應用，從而在很多天然化合物，農藥醫藥，新興材料以及納米分子器件的合成中起著關鍵的作用。

反應定義

在Pd/Cu催化下，末端炔與芳基或烯基鹵代物的交叉偶聯反應。此反應最好的鈀催化劑是Pd(PPh3)2Cl2或Pd(PPh3)4，而且反應是化學定向的，反應前后底物和產品的化學構型不變。


反應特征

1)Sonogashira偶聯通常可以在室溫或略高于室溫的溫度下進行，必須嚴格控制無氧；

2)使用催化用量的銅(I)鹽來避免處理沖擊敏感/爆炸性的銅乙炔；

3)銅(I)鹽可以是市面上可以買到的CuI或CuBr；

4)通常堿作為溶劑，但偶爾也使用助溶劑;

反應實例

1. 在Pd-NHC催化下末端炔烴與芳基三甲基三酸銨發生交叉偶聯反應。反應條件溫和，炔烴底物耐受烷基、芳基和雜芳基等多種官能團。反應過程中胺可以通過一鍋法直接烷基化，無需分離銨鹽[1]。

2.該方法使用非貴金屬催化劑選擇性C(sp2)-C(sp)交叉偶聯，即利用可見光催化Cu催化Sonogashira型反應，可以從不同芳烴衍生的芳基磺酸鹽中選擇性烷基化合成高選擇性和高官能團相容性的芳基炔[2]。
